If you use Gnip you can specify up to 29 keywords per rule. The current allotment of Gnip rules per user will allow you to more than cover 500 keywords. The cost in terms of credits ($3/10,000) will depend on the frequency of the keywords.
DiscoverText also provides free access to the Twitter Search API. No programming skills are required, but you do need a valid Twitter account to use this service.
